CDg13 is a fluorescent probe that specifically detects living neural stem/progenitor cells. CDg13 localizes to the endoplasmic reticulum via its dihexyl moiety, with no interaction with endoplasmic reticulum biomolecules. CDg13 undergoes selective efflux by active ABCG2 transporters, with increased intracellular accumulation following ABCG2 inhibition or knockdown. CDg13 isolates and enriches self-renewable neural stem/progenitor cells from embryonic mouse brain tissue, stains neural stem/progenitor cells brightly, and sorts ABCG2low cell populations from heterogeneous populations. (Ex/Em = 520/553 nm)

Description

CDg13 is a fluorescent probe that specifically detects living neural stem/progenitor cells. CDg13 localizes to the endoplasmic reticulum via its dihexyl moiety, with no interaction with endoplasmic reticulum biomolecules. CDg13 undergoes selective efflux by active ABCG2 transporters, with increased intracellular accumulation following ABCG2 inhibition or knockdown. CDg13 isolates and enriches self-renewable neural stem/progenitor cells from embryonic mouse brain tissue, stains neural stem/progenitor cells brightly, and sorts ABCG2low cell populations from heterogeneous populations[1]. In Vitro

CDg13 (1 μM; 1 h) selectively stains live mouse NS5 (NSPC) cells with bright fluorescence, while showing minimal staining of MEF, mESC, and differentiated NS5 cells[1].
CDg13 (1 μM; 1 h) fluorescence in D-NS5 cells is inversely correlated with Abcg2 expression, as demonstrated by enhanced fluorescence in Abcg2-knockdown cells stained with it[1].
CDg13 (1-10 μM; 48 h-7days) has very low toxicity to live mouse NSPCs[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Immunofluorescence[1]

Cell Line: mouse embryonic fibroblasts (MEF), mouse embryonic stem cells (mESC), mouse NSPC line (NS5), differentiated NS5 (D-NS5)
Concentration: 1 μM
Incubation Time: 1 h
Result: Showed bright fluorescence in NS5 cells, with significantly lower fluorescence in MEF, mESC, and D-NS5 cells, as visualized by fluorescence microscopy and confirmed by flow cytometry histograms.

Cell Cytotoxicity Assay[1]

Cell Line: mouse NSPCs (NS5 cells)
Concentration: 1, 10 μM
Incubation Time: 48 h; 7 days;
Result: Showed no cell toxicity after incubation with 10 μM for 48 h.
Showed no effect on neurosphere size after incubation with 1 μM for 7 days.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
