DEC-RVRK-CMK (Decanoyl-Arg-Val-Arg-Lys-chloromethylketone) is a peptidyl CMK inhibitor that targets the calcium-dependent subtilisin-like proprotein convertase family. DEC-RVRK-CMK is a furin-targeting proprotein convertase inhibitor that blocks the cleavage and maturation of inhibin α-subunit and βB-subunit precursors. DEC-RVRK-CMK has no inhibitory effect on the cleavage conversion of ADAM2 protein from 46 kDa to 27 kDa during sperm capacitation, whereas the broad-spectrum kexin-like convertase inhibitor dec-RVKR-CMK (HY-107760) can block this processing, indicating that this cleavage is mediated by other members of the PCSK family such as PCSK7 and is independent of PCSK4^[1].
DEC-RVRK-CMK (RVRK) (10 μM; 24-48 h) does not inhibit branching morphogenesis of E15 rat SMG rudiments in organ culture or reduce AQP5 expression^[4].
DEC-RVRK-CMK (1 h) blocks proconvertase-mediated cleavage of inhibin α- and βB-subunits in LβT2 pituitary gonadotrope cells, leading to accumulation of unprocessed pro-αN-αC and pro-βB^[2].
DEC-RVRK-CMK inhibits purified recombinant rat PC4 (rec-rPC4-FLV5 E1-pool) with an IC₅₀ of 12.2 nM^[3].
