MTS peptide
MTS peptide is a short peptide with a mitochondria-targeting sequence. In F-M-LNP, MTS peptide is displayed on the particle surface via thiol-maleimide conjugation, and is specifically responsible for being recognized by the mitochondrial proteins TOMM20/22, "anchoring" the nanoparticles to the outer mitochondrial membrane, followed by membrane potential-independent bilayer membrane penetration mediated by fluorinated lipids. MTS peptide can be used in the research of Leber's hereditary optic neuropathy.

Biologische Aktivität
MTS peptide modification enhances the mitochondrial targeting selectivity of 6F-LNPs in N2a cells by increasing interactions with TOMM20 and TOMM22, resulting in a Pearson's colocalization coefficient of 0.90 between delivered pDNA and mitochondria[1].
MTS peptide-modified 6F-M-LNPs (2 μg/mL; 24 h followed by 48 h fresh medium culture) deliver functional hND4 to GM10742 LHON patient cells, restoring mitochondrial function with a 5-fold increase in hND4 protein expression and significant improvements in MMP, ATP production, and oxygen consumption[1].
