Integrin beta 6

Integrin beta 6, the ITGB6 gene product, is an epithelium-specific beta subunit known to pair only with alpha V. The resulting alpha V beta 6 heterodimer activates latent TGF-beta-1 and TGF-beta-3 by binding the RGD motif of the latency-associated peptide, coupling integrin ligation to cytokine release that governs wound healing, mucosal immunity and fibrosis. Beta-6 is virtually absent from resting adult airway epithelium but strongly induced during injury and in idiopathic pulmonary fibrosis, providing the rationale for current anti-beta-6 clinical programs. Loss-of-function variants in ITGB6 cause hypomaturation amelogenesis imperfecta affecting dental enamel, and the subunit is overexpressed in several carcinomas as a poor-prognosis marker.
