PIM2

Located on the X chromosome, PIM2 is the paralog most abundant in brain, hematopoietic cells and testis, and it is strongly upregulated in chronic lymphocytic leukemia, mantle cell lymphoma and multiple myeloma. Like PIM1, it is a constitutively active kinase controlled at the level of expression and stability, and it phosphorylates substrates such as BAD, 4E-BP1 and eIF4B to promote survival and cap-dependent translation. In the nervous system PIM2 can compensate for PIM1 loss, and Pim1/Pim2 double-knockout mice show growth retardation. Its restricted expression pattern makes PIM2 a focus of isoform-selective inhibitor development.
