Thionin Staining Solution (1%) is mainly used for chromosome staining, such as somatic chromosome and meiosis staining, in which metachromatic substances appear purplish red, while other substances appear blue.

Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
1. Cell Sample Staining
1.1 Collect cell samples, centrifuge at 1500 g for 10–15 min, and discard the supernatant.
1.2 Add fresh fixative (methanol:glacial acetic acid = 3:1), mix gently, and incubate at room temperature for 30 min.
1.3 Centrifuge again at 1500 g for 10–15 min and discard the supernatant.
1.4 Depending on the cell quantity, add a few drops of freshly prepared fixative and mix thoroughly to form a suspension.
1.5 Place 1 drop of the suspension onto a clean, pre-cooled microscope slide and allow it to air-dry.
1.6 Staining results: Metachromatic substances appear purplish-red; others appear blue.
2. Chromatin Staining
2.1 Place the slide specimen in 1 M HCl and incubate at 37°C for 20 min.
2.2 Rinse thoroughly with distilled water and allow to air-dry.
2.3 Immerse the slide specimen in thionine staining solution (0.4%) for 15–20 min.
2.4 Rinse with distilled water and allow to air-dry.
2.5 Observe under a microscope.
2.6 Staining results: Metachromatic substances appear purplish-red; others appear blue.
