In Vitro
The structure of DOPE-mPEG comprises two key components that work synergistically:
DOPE: Containing two oleoyl groups (C18:1), it imparts excellent membrane fluidity and promotes membrane fusion under acidic conditions (such as endosome escape).
PEG chain: Acts as a hydrophilic spacer arm, forming a hydration layer on the particle surface, reducing non-specific adsorption and immune system recognition and clearance, thus prolonging the carrier's circulation time in vivo.
