Hadacidin
Based on 1 Customer Validation
Hadacidin is a competitive inhibitor of adenylate synthase, possessing antitumor, plant growth inhibitory, and other activities. Hadacidin competitively inhibits aspartate binding, blocking the formation of adenylosuccinate, the reamination of IMP, and de novo adenine nucleotide synthesis. Hadacidin inhibits mitosis and plant stem/root growth. Hadacidin induces mesenchymal/epithelial cell death at high concentrations. Hadacidin can be used in plant growth regulation and cancer-related research.

Biological Activity
Description
In Vitro
Hadacidin (2-20 mg/mL; 2-4 days) inhibits morphogenesis and mitosis in organ cultures of mandibular molar tooth germs of C57BL/6J mice; the effect is reversible after transfer to control medium; administration of adenine (0.05 mg/mL) or adenosine (0.1 mg/mL) partially prevents this effect[1].
Hadacidin competitively inhibits D. discoideum Ax-3 AMPS synthetase, producing approximately 50% inhibition at 100 μM and >95% inhibition at approximately 250 μM, with a Ki of 1.5 × 10-6 M[5].
Hadacidin competitively inhibits adenylosuccinate synthetase by competing with L-aspartate, blocks adenylosuccinate formation, and inhibits de novo biosynthesis of adenylate/deoxyadenylate in in vitro assays[6].
Hadacidin (5 mg/mL; 12-18 hours) reduces aspartate and thymidine uptake in D. discoideum Ax-3 by at least 75%; increases osmotic stability of D. discoideum Ax-3[5].
Hadacidin prevents the growth of D. discoideum Ax-3 in FM and HL-5, and growth resumes after washing out[5].

In Vivo
Hadacidin inhibits stem and root growth of Alaska pea plants grown in nutrient solution, and 16 days after foliar spraying, the average stem length decreases from 352 mm at 0 mg/L to 96 mm at 8000 mg/L[2].
Hadacidin inhibits the unfolding of the first trifoliolate leaf of pinto bean; when applied to the primary leaves of pinto bean, it is transported to the young trifoliolate leaves and inhibits their growth. In the bean leaf bioassay, the inhibition is counteracted by riboflavin, but not by glycine, whereas aspartic acid, asparagine, glutathione, and calcium pantothenate partially reverse it[2].
Hadacidin applied to tomato roots in nutrient solution completely stops root growth at 50 and 250 mg/L, and inhibits shoot growth by 40% at 50 mg/L and completely at 250 mg/L[2].
Hadacidin (100 mg/kg; i.v.; 30 min before stimulation) effectively blocks IMP (HY-108213) reamination in fast muscle of recovered rats; it also increases IMP accumulation in fast-twitch white gastrocnemius muscle, with no significant effect on IMP levels in fast-twitch red gastrocnemius muscle[3].

In Vivo:
Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (20.99 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (20.99 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
