Description

Clofexamide (ANP 246) is an orally active antidepressant agent, exhibiting analgesic and anti-inflammatory activity. Clofexamide can be used for the research of depression, inflammatory disorders, peptic ulcer[1][2].

In Vivo

Clofexamide (200 mg/kg; p.o.; single dose) is rapidly excreted in urine (approx. 85% within 24 h) via N-deethylation, hydroxylation, amide hydrolysis, N-oxidation, and glucuronidation in rats[2].

Animal Model: Wistar rats (male; 300-400 g, 8-week-old)[2]
Dosage: 200 mg/kg
Administration: p.o.; single dose
Result: Excreted approximately 85% of the dosed clofexamide in urine within 24 hours, with 36% as unchanged clofexamide, 20% as 2-(p-chlorophenoxy)-N-(2-ethylaminoethyl) acetamide, 7% as 2-(p-chlorophenoxy)-N-(2-aminoethyl) acetamide, 7% as 2-(4-chloro-2-hydroxyphenoxy)-N-(2-diethylaminoethyl) acetamide, 15% as p-chlorophenoxyacetic acid, plus minor amounts of 2-(p-chlorophenoxy)-N-(2-diethylaminoethyl) acetamide N-oxide and p-chlorophenoxyaceturic acid.
Detected additional urinary metabolites including 2-(4-chloro-3-hydroxyphenoxy)-N-(2-diethylaminoethyl) acetamide, 2-(4-chloro-3-hydroxyphenoxy)-N-(2-ethylaminoethyl) acetamide, 2-(4-chloro-2-hydroxyphenoxy)-N-(2-ethylaminoethyl) acetamide.
Detected only unchanged clofexamide in feces.
