Cy3-N3 is a Cy3-azide fluorescent dye used to label for protein and nucleic acid. Cy3-N3 is a click chemistry reagent, it contains an Azide group and can undergo copper-catalyzed azide-alkyne cycloaddition reaction (CuAAc) with molecules containing Alkyne groups. It can also undergo strain-promoted alkyne-azide cycloaddition (SPAAC) reactions with molecules containing DBCO or BCN groups (Ex/Em = 548/563 nm).

Description

Cy3-N3 is a Cy3-azide fluorescent dye used to label for protein and nucleic acid. Cy3-N3 is a click chemistry reagent, it contains an Azide group and can undergo copper-catalyzed azide-alkyne cycloaddition reaction (CuAAc) with molecules containing Alkyne groups. It can also undergo strain-promoted alkyne-azide cycloaddition (SPAAC) reactions with molecules containing DBCO or BCN groups (Ex/Em = 548/563 nm).

In Vitro

Guidelines (The following is our recommended protocol; this is a guideline and should be modified according to your specific needs).
1. Dye Preparation
1.1 Preparation of Stock Solution Prepare a 25 mM stock solution using anhydrous DMSO.
Note: It is recommended to aliquot the stock solution and store it at -20 ℃ or -80 ℃ protected from light.
1.2 Preparation of Working Solution Dilute the stock solution with preheated PBS to prepare a 25 μM working solution.
Note: Please adjust the working solution concentration according to your actual situation, and prepare it fresh before use.
2. Click Reaction
2.1 Collect cells and lyse them, retaining the protein in the cell lysate (1 mg/mL).
2.2 Add Cy5-N3 (final concentration 25 μM), TCEP (final concentration 1 mM), and TBTA (final concentration 100 μM).
2.3 Vortex mix
2.4 Add CuSO4 (final concentration 1 mM) and incubate by rotation at room temperature for 1-2 hours.
2.5 Precipitate using a protein precipitation kit to remove excess click reaction reagent.
2.6 Separate and detect by gel electrophoresis.
