DPPC (129Y83) is a phosphoglyceride that can be used to prepare lipid monolayers, bilayers, and liposomes. DPPC is the main lipid component of pulmonary surfactant. Dppc-liposome can be effectively used as a delivery vector to induce an immune response against GSL antigen in mice.

DPPC (2-10 mg/ml) monolayer can be inserted by Aβ40, but the amount of insertion is smaller than that of DPPG or DPTAP. The behavior of Aβ40 inserted into the DPPC monolayer is affected by surface pressure, solvent (water or PBS), and salt concentration[3]. DPPC can mix with DPPE to form DPPC/DPPE lipid bilayer systems. As DPPE concentration rises, the average area per headgroup diminishes (0.65 to 0.52 nm²), enhancing membrane compactness. Meanwhile, membrane thickness slightly increases (3.43 to 4.00 nm), reflecting lipid tail ordering. [5].

This protocol yields a suspended solution of 2.5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.
Taking 1 mL working solution as an example, add 100 μL EtOH stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
