Dns–LLC
Dns-LLC is a cell-permeable Cu+-selective fluorescent probe that forms a 1:1 complex with Cu+. The thiol group of Dns-LLC plays a key binding role, while the sulfonamide and amide groups jointly contribute to the stabilization of the complex. Upon binding to Cu+ in aqueous buffer solutions, Dns-LLC generates a turn-on fluorescence response, which can be used for the detection of Cu+ in the Golgi apparatus.

Biological Activity
Dns-LLC (10 μM) forms a 1:1 complex with Cu+ in buffered aqueous solution at physiological pH, exhibits approximately 12-fold fluorescence turn-on enhancement towards Cu+, and has a femtomolar-level binding affinity (Kd=12 fM)[1].
Dns-LLC (20 μM; 30 min) penetrates live human colorectal RKO cells, selectively detects intracellular Cu+ via a turn-on fluorescence response, localizes to the Golgi apparatus to detect Cu+ in this organelle, and exhibits no obvious cytotoxicity against RKO cells[1].
Dns-LLC holds potential for the study of cuproptosis.
