DSG-PEG2000-WYRGRL is a functionalized PEGylated lipid conjugate composed of three modular components: distearoylglycerol (DSG) as the lipid anchoring group, a polyethylene glycol (PEG) spacer arm with a molecular weight of 2000 Da, and WYRGRL peptide (HY-P10739)as the terminal targeting/functional ligand. Biological Activity
DSG-PEG2000-WYRGRL is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against specific cell surface receptors in tumor and inflammatory microenvironments. The 2000 Da PEG chain ensures prolonged blood circulation time and immune evasion, while the DSG anchoring group provides stable integration into the nanoparticle membrane.
Composition and role of each component:
1. DSG (distearoylglycerol): Lipid anchoring group-a diacylglycerol with two saturated C18 chains; provides strong membrane rigidity and sustained nanoparticle stability, suitable for applications requiring prolonged circulation time.
2. PEG2000 (polyethylene glycol, MW 2000): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
3. WYRGRL (WYRGRL peptide): Targeting ligand-a targeting peptide with the sequence tryptophan-tyrosine-arginine-glycine-arginine-leucine, designed to enhance tissue-specific binding. The WYRGRL motif has been explored for targeting specific cell populations in tumor and inflammatory microenvironments, utilizing its arginine-rich sequence to simultaneously achieve cell penetration and targeting functions.
