Description

EB1 peptide (Penetrating analog) is an endosomolytic agent and siRNA delivery agent. EB1 peptide forms an amphipathic alpha helix upon protonation in early-late endosomes, drives endosomal membrane permeabilization, and enables endocytosed siRNA escape into the cytosol. In Vitro

EB1 peptide (10:1-25:1 molar ratio of EB1 peptide: siRNA; 1 h) delivers significantly more siRNA into HeLa cells at low molar ratios, with 10-fold higher uptake at 10:1 and 2.5-fold higher uptake at 25:1 compared to Penetratin (HY-P2529)[1].
EB1 peptide (25:1 molar ratio of EB1 peptide: siRNA; 36 h) delivers biologically active siRNA to transiently transfected HeLa cells, significantly reducing luciferase expression to ~50% of control at 100 nM siRNA in both serum-free and serum-containing conditions, and to ~70% of control at 10 nM siRNA in serum-free conditions[1].
EB1 peptide (25:1 molar ratio of EB1 peptide: siRNA; 48 h) delivers biologically active siRNA to stably transfected HepG2 cells when preincubated in a large volume, significantly reducing luciferase expression to ~65% of control at 100 nM siRNA after 48 h[1].
EB1 peptide (25:1 molar ratio of EB1 peptide: siRNA; 36 h) promotes more effective endosomal escape of siRNA in transiently transfected HeLa cells than HA2-penetratin-augmented Penetratin, resulting in greater luciferase gene silencing at a 25:1 peptide: siRNA ratio after 36 h[1].
