Biological Activity
This product can be used for: protein enzyme digestion sequencing analysis, peptide map analysis and peptide mass fingerprint analysis.
Reaction conditions
Reaction pH: active between 4.0-10.0
Optimum pH: 8.0-8.5
Inhibitors: Diisopropylfluorophosphate (DFP), α2-macroglobulin, and Nα-P-tosyl-L-lysine chloromethyl ketone (TLCK)
Molecular weight: 24.0±2.4 kDa (SDS-PAGE)
Protocol
1)Dissolution: 25-50 mM NH4HCO3 (pH 7.8) to dissolve the target protein. If the solubility is not good, the target protein can be denatured (adding urea, SDS, DTT or heating).
2)Enzyme digestion: The recommended ratio of V8 protease and target protein is 1/20-1/100 (W/W), enzyme digestion at 25°C or 37°C for 2-18 h.
