Estrogen receptor modulator 8
Estrogen receptor modulator 8 (Compound 4) down-regulates estrogen receptor with an IC50 of 0.1 nM-10 nM. Estrogen receptor modulator 8 degrades ERα (IC50: 25.7 nM in vitro, 0.136 nM in MCF-7 cell). Estrogen receptor modulator 8 also inhibits MCF-7 cell growth (IC50: 0.1 nM), and MCF-7 tumor growth.

Estrogen receptor modulators are compounds that selectively regulate the activity of estrogen receptors (ERα/ERβ). They can exhibit estrogen agonist or antagonist effects in different tissues (i.e., tissue selectivity). For example, tamoxifen exhibits anti-estrogen effects in the breast but estrogen-like effects in bone, and is used for breast cancer prevention and osteoporosis treatment.
