Description

FD-1080 is a fluorophore with both excitation and emission in the NIR-II region (Ex=1064 nm, Em=1080 nm). FD-1080 can be used for in vivo imaging[1].

In Vitro

Guide (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
Dilute the stock solution in serum-free cell culture medium or PBS to obtain 1-10 μM of working solution.
1. Preparation of FD-1080 Working Solution
1.1 Preparation of Stock Solution
Dilute FD-1080 with DMSO to prepare a 10 mM FD-1080 stock solution.
Note: It is recommended to aliquot the FD-1080 stock solution and store it at -20°C or -80°C, protected from light.
1.2 Preparation of Working Solution
Dilute the stock solution with pre-warmed PBS to prepare a 2–10 μM FD-1080 working solution. Sterilize the solution by filtration through a 0.2 μm filter membrane.
Note: Please adjust the concentration of the FD-1080 working solution according to specific experimental requirements; prepare fresh solution immediately before use.
2. Cell-based Experiments
2.1 Suspension Cells: Centrifuge to collect the cells, then add PBS and wash twice (5 minutes per wash).
Adherent Cells: Discard the culture medium, then add trypsin to detach the cells. After centrifuging to remove the supernatant, add PBS and wash twice (5 minutes per wash).
2.2 Add 1 mL of the FD-1080 working solution and incubate at room temperature for 10–30 minutes.
2.3 Centrifuge at 400 g at 4°C for 3–4 minutes, then discard the supernatant.
2.4 Add PBS and wash the cells twice (5 minutes per wash).
2.5 Resuspend the cells in 1 mL of serum-free culture medium or PBS, then analyze using a fluorescence microscope or flow cytometer.
3. In Vivo Injection
3.1 Intravenously inject 200 μL of the 80 μM FD-1080 working solution into a mouse; perform in vivo imaging analysis 10–20 minutes after injection.

In Vivo

The 1064 nm NIR-II excitation of FD-1080 is demonstrated with the high tissue penetration depth and superior imaging resolution compared to NIR excitation from 650 nm to 980 nm. Deeptissue and high-resolution in vivo imaging for the left hindlimb vasculature, abdomen, and brain vessels was realized, allowing penetration through intact skin, tissue, and skull. FD-1080 also quantifying the respiratory rate based on the dynamic imaging of respiratory craniocaudal motion of the liver for the awake and anaesthetized mouse[1].
