Bioluminescence imaging usually measures luciferase activity after substrate administration and is commonly used as a surrogate for viable reporter-expressing tumor burden, while fluorescence imaging measures reporter or probe emission and can support tumor localization, ex vivo confirmation, or complementary multimodal analysis.
