In Vitro

GIGA-564 (0.001-100 μg/mL) exhibits extremely low activity in blocking the binding of CTLA-4 to CD80 and CD86 in ELISA assays[1].

In Vivo

GIGA-564 (5 mg/kg; i.p.; once every 3 days; 3 doses) induces a lower level of peripheral Treg proliferation than Ipilimumab (HY-P9901) in hCTLA-4 KI mice bearing MC38 tumors[1].
GIGA-564 (0.3 mg/kg; i.p.; once every 3 days; 3 doses) exhibits excellent anti-tumor activity in hCTLA-4 KI mice bearing MC38 tumors[1].
GIGA-564 (30 mg/kg; i.p.; once every 3 days; 9 doses) induces less toxicity than Ipilimumab (HY-P9901) in hCTLA-4 knock-in (KI) and hCTLA-4/hPD-1 double knock-in (KI) mice, as evidenced by lower liver enzyme levels, reduced colitis, and the absence of skin lesions at the tested dose[1].
