GluN1 (356-385) is a polypeptide targeting NMDAR GluN1. GluN1 (356-385) induces the production of autoantibodies, which reduce the density of cell surface NMDAR clusters, impair long-term potentiation, and decrease NMDAR-mediated Ca2+ influx. As an immunogen, GluN1 (356-385) induces symptoms similar to anti-NMDAR encephalitis, including memory loss, in mice. GluN1 (356-385) can be used to establish a mouse model that mimics the pathogenesis of anti-NMDAR encephalitis. GluN1 (356-385) is applicable to research related to anti-NMDAR encephalitis.

In Vitro
GluN1(356-385)-induced antibody (24 h) reduce the density of surface GluN1-positive synaptic clusters and decrease membrane GluN1 levels in primary hippocampal neurons from embryonic day 18 rats[1].
GluN1(356-385)-induced antibody (18 h at 37 °C) significantly reduces NMDAR-mediated calcium influx in primary rat hippocampal neurons on embryonic day 18[1].

Animal Model:C57BL/6 (female, 10 weeks old, induced by active immunization with GluN1(356-385) peptide)[1]

Dosage:200 μg

Administration:s.c.; initial immunization on back, followed by two booster injections at 4 and 8 weeks after initial immunization

Result:Showed significant memory loss, with decreased exploration time of a novel object and a lower discrimination index in the novel object recognition test .
Reduced visits to strangers in the three-chamber test.
Reduced hippocampal membrane GluN1 levels.
Impaired long-term potentiation (LTP) at Schaffer collateral-CA1 synapses, with a normalized field excitatory postsynaptic potential slope of 112% compared to 143% in controls.
Induced autoantibodies targeting GluN1 in cerebrospinal fluid and serum, maintained at high titers through the 12-week period, and localized primarily to the hippocampus and cerebellum.
