Glutamate oxidase (L-Glutamate oxidase) is a FAD-dependent oxidase with high substrate selectivity. Glutamate oxidase catalyzes the oxidative deamination of L-glutamate using O2 to produce α-ketoglutarate (α-KG), NH3, and H2O2. Glutamate oxidase is applicable to research related to the biocatalytic production of α-KG and the analytical detection of L-glutamate.

Biological Activity
Glutamate oxidase (L-Glutamate oxidase) selectively catalyzes the conversion of L-glutamate into α-ketoglutarate, NH3 and H2O2. The L-glutamate oxidase derived from Streptomyces sp. X-119-6 has a Km value of 0.21 mM for L-glutamate, and its activity toward L-aspartate is merely 0.6% of that toward L-glutamate[2].
Glutamate oxidase (1.5 U/mL; 24 h) produces 104.7 g/L α-KG with a conversion rate of 95.8% under the conditions of 250 U/mL catalase, 3 mM MnCl2, 110 g/L L-glutamate, 30 °C and pH 6.5[4].
Glutamate oxidase only converts L-glutamate among the 25 tested amino acids. The Km values of L-glutamate oxidase derived from Streptomyces endus for L-glutamate and O2 are 1.1 mM and 1.86 mM, respectively[3].
