Glycolyl-CoA
Glycolyl-CoA is a carboxyl acceptor substrate and a FORCE/TaCo pathway metabolite, can be generated by HACL-mediated condensation of formyl-CoA with formaldehyde, and can also be formed from glycolate via glycolyl-CoA synthetase, is carboxylated by GCC to tartronyl-CoA and supports the tartronyl-CoA photorespiration bypass for carbon fixation, and simultaneously can serve as an intermediate in HB-5.2-catalyzed oxidative condensation of ethylene glycol. Glycolyl-CoA can be used for research on metabolic pathways and carbon fixation mechanisms.

Biological Activity
Description
In Vitro
Glycolyl-CoA (0.5 mM; preincubated at 37°C for 2 min) can be carboxylated by purified GCC M5, G20R, and L100N, among which G20R has the highest kcat (9.8 s−1) and GCC M5 has the lowest apparent KM (0.15 mM)[5].
Glycolyl-CoA (0.5 mM; preincubated at 37°C for 2 min) as substrate: L100N consumes 1.7 ATP per carboxylation, whereas GCC M5 consumes 4.0[5].
Glycolyl-CoA (injection volume 1 μL; flow rate 0.25 mL/min; column temperature 25°C) is quantified by LC-MS/MS in positive ion mode using the precursor ion 826.1 and product ions 428.1/319.1[5].
Glycolyl-CoA (4 mM; blotting 3.5 s) enables the cryo-EM structures of GCC M5 G20R and L100N at resolutions of 2.05 and 2.31 Å, respectively[5].
