DPPD (8-300 mg/kg bw/day; p.o.; daily; 42 days (males); 42-46 days (females)) significantly prolongs gestation length at doses of 50 and 300 mg/kg bw/day in female [Crl:CD(SD)] SPF rats, with the reproductive/developmental NOAEL established at 8 mg/kg bw/day[1].

Result:Decreased food consumption significantly in the fourth week at 300 and 1000 mg/kg bw/day in male rats.

Increased total bilirubin levels significantly at all doses in male rats at the end of dosing period, which resolved during recovery.
Decreased γ-GTP to 0.63 IU/L at 1000 mg/kg bw/day in female rats, within normal background range.
Showed no significant dose-related effects in clinical signs, sensory/motor function, body weight, hematology, urine parameters, organ weights, or histopathology.

Prolonged gestation length significantly at 50 and 300 mg/kg bw/day (mean 23.0 days vs control 22.4 days).
Showed non-significant dose-dependent decrease in number of pups born, delivery index, number of live pups, birth index, live birth index (day 0 of lactation), number of live pups, and viability index (day 4 of lactation), most marked at 300 mg/kg bw/day.
Showed no effects in male fertility, estrous cycle length, implantation indices, pup body weights, or pup malformations.
