PI-16

PI-16 is a selective uroporphyrinogen decarboxylase (UROD) inhibitor with an IC₅₀ of 9.9 µM against human UROD. PI-16 inhibits the heme biosynthesis enzyme that converts uroporphyrinogen to coproporphyrinogen. PI-16 selectively reduces the viability of hypopharyngeal squamous cell carcinoma and cervical cancer cells. PI-16 can be used in combination with Cisplatin (HY-17394), with synergistic effects observed in hypopharyngeal squamous cell carcinoma cells. PI-16 can be used in research related to hypopharyngeal squamous cell carcinoma and cervical cancer.

Others

PI-16 (62.5 mM; 35 min-1 h) inhibits recombinant human UROD with an IC₅₀ of 9.9 μM but does not inhibit PBGD at 62.5 mM, demonstrating specificity in cell-free heme biosynthesis enzyme assays^[1].
PI-16 (24 h) combined with cisplatin is potently synergistic in FaDu cells (CI<1), and PI-16 with radiation or cisplatin shows at-least-additive/more-than-additive effects in FaDu and ME-180 clonogenic assays^[1].

Anti-cancer

PI-16 reduces viability of FaDu and ME-180 cells with EC₅₀ values of 22.7 μM and 26.9 μM, respectively, while normal NOE cells show EC₅₀ >50 μM^[1].
