CGC-11150

CGC-11150 is a decamine-based polyamine analog that acts as a voltage-dependent pore blocker of Kir channels. CGC-11150 produces voltage-dependent pore block similar to that of Spermine (HY-B1777) in recombinant Kir6.2^{[N160D, L164C, C166S, Δ36]} + SUR1 channels, but with faster on and off rates. CGC-11150 inhibits the development of and induces regression of laser-induced choroidal neovascularization (CNV). CGC-11150 is used in studies related to choroidal neovascularization.

In Vitro

CGC-11150 (1 μM) induces voltage-dependent inward rectification of Kir6.2-rec channels expressed in COSm6 cells, with blocking characteristics similar to spermine but slightly reduced potency and faster on/off rates^[1].

In Vivo

CGC-11150 (10-20 mg/kg; i.p.; 2 to 3 times per week; for 14 days) causes a slight reduction in CNV area, but exhibits poor tolerability at high doses^[2].
CGC-11150 (20 μg; intravitreal injection; administered on day 0 and day 7 post-injury) reduces the CNV area by approximately 40% in a mouse model of laser-induced choroidal neovascularization^[2].

Animal Model:

C57BL/6J (female, 4 to 5 weeks old, laser-induced rupture of Bruch’s membrane)^[2]
Dosage:

10 mg/kg; 20 mg/kg (i.p.); 20 μg (intravitreous)
Administration:

i.p.; twice a week; 14 days; i.p.; three times a week; 14 days; intravitreous; on days 0 and 7 after injury
Result:

Caused a small reduction in CNV lesion area.
Was not well tolerated (several mice died), but surviving mice showed a small reduction in CNV lesion area comparable to the 10 mg/kg dose.
Resulted in an approximately 40% reduction in CNV lesion area compared to vehicle-treated or untreated eyes.
Caused retinal damage.
