Misitatug blivedotin (RC88) is an antibody-drug conjugate (ADC) targeting mesothelin (MSLN). Misitatug blivedotin is formed by conjugating the humanized anti-mesothelin monoclonal antibody Misitatug (HY-P990929) with the microtubule inhibitor Monomethyl auristatin E (MMAE) (HY-15162) via a cleavable linker. Misitatug blivedotin binds to mesothelin and exhibits dose‐dependent antitumor activity. Misitatug blivedotin can be used for the research of ovarian cancer, non-squamous non-small-cell lung carcinoma, cervical cancer.

Animal Model:Balb/c nude mice (female; ages 4–5 weeks; implanted subcutaneously with 3 million OVCAR3‐MSLN‐3# or H292‐MSLN‐9C8 cells for a xenograft model)[3]

Dosage:0.7 mg/kg; 0.75 mg/kg; 1.5 mg/kg; 2.1 mg/kg; 3 mg/kg

Result:Had moderate activity at 0.75 mg/kg with single dosing, but tumor volume was less than the initial volume of tumors treated at 2.1 mg/kg until 11 days after dosing.
Significantly reduced the tumors 7 days after the first dosing at 1.5 and 3 mg/kg with multiple dosing.
Fully regressed tumors at 3 mg/kg for 10 days and at 1.5 mg/kg for 17 days, but the tumor continued to grow after three administrations at 0.75 mg/kg.

Subcutaneous Cell-Line-Derived Xenograft
Subcutaneous cell-line-derived xenograft (CDX) models are established by implanting cultured human cancer cell lines into immunodeficient mice, where the injected cells form localized tumors that can be monitored in vivo as a measure of tumorigenic potential, growth kinetics, and treatment response. These models are widely used in oncology research because they allow reproducible tumor formation and enable comparative assessment of tumor growth between different cell lines or genetic manipulations in a controlled in vivo microenvironment. Subcutaneous implantation of cancer cells in immunodeficient mice is a standard approach for evaluating tumor growth behavior and therapeutic response across multiple cancer types, including prostate, esophageal, pancreatic, and colon cancer models.
