Nelutroctiv activates cardiac sarcomeres by enhancing the sensitivity of troponin to calcium ions and promoting the interaction between actin and myosin. Nelutroctiv enhances cardiac contractility. Nelutroctiv can be used in the researches for cardiovascular diseases with impaired cardiac contractile function.

In Vitro

Nelutroctiv (1 μM) reduces the calcium concentration for 50% maximum tension (EC₅₀) by 16%-25% and increases maximum tension (T_max) by 16%-50% in permeabilized myocytes from human HFrEF, HFpEF (with or without severe obesity)^[1].

In Vivo

Nelutroctiv (0.5 mg/kg i.v.; 2.0 mg/kg p.o.) shows low clearance (3.8 mL/min/kg) and high oral bioavailability (84%), with concentration-dependent increases in fractional shortening in Sprague-Dawley rats^[2].
