Description

MeDZ lipid is a zwitterion-type ionizable endosomal membrane destabilizer and anti-inflammatory agent that promotes endosomal escape. When incorporated into LNP formulations, MeDZ lipid enhances mRNA expression in lymph node antigen-presenting cells and promotes cytotoxic T cell activation. MeDZ lipid is compatible with existing targeted nanoparticle formulations to improve mRNA delivery efficiency[1].

In Vitro

MeDZ lipid (10 min; pH 7.4-5.5) exhibits pH-responsive protonation, transitioning from a zwitterionic state with bound H2O at pH ≥6.8 to a positively charged state with bound H3O+ at pH <6.8, as detected via 1H NMR spectroscopy[1].
LNPs containing 25 mol% MeDZ lipid (1 μg/mL mRNA; 3 h incubation, 24 h expression incubation) maximize EGFP mRNA expression in DC2.4 cells, with 1.7-fold higher expression than control LNPs despite equivalent cellular uptake[1].
LNPs containing 25 mol% MeDZ lipid (pH 6.0) exhibit enhanced membrane disruption activity at acidic pH (6.0) compared to control LNPs, supporting improved endosomal escape capability[1].
LNPs containing 25 mol% MeDZ lipid (15 μg/mL; 1.5 h incubation) exhibit enhanced endosomal escape in DC2.4 cells, as shown by reduced co-localization with endosomes/lysosomes (PCC = 0.78) compared to control LNPs (PCC = 0.90)[1].
LNPs containing 25 mol% MeDZ lipid (1 μg/mL OVA mRNA; 6 h incubation, 24 h expression/presentation incubation) enhance OVA antigen expression and presentation in BMDCs, with OVA protein levels at ~100% of control LNPs and 16.0% H2Kb-SIINFEKL-positive cells post-treatment[1].
