mPEG2000-CM (mPEG2000-COOH) is a carboxyl-terminated monomethoxy polyethylene glycol. mPEG2000-CM bears a reactive carboxyl group (-COOH) at its structural terminal site, which can form stable amide bonds with amino groups and ester bonds with hydroxyl groups. mPEG2000-CM binds to PCA-g-PCL copolymers via electrostatic interaction to form polyion complex micelles with a hydrophilic PEG surface, which enhances the stability of micelles in aqueous media. mPEG2000-CM can be used for drug delivery[1][2].

In Vitro

mPEG-CM (MW 2000) (5 h at 150 °C) is successfully synthesized by reacting poly(ethylene glycol) methyl ether (Mn = 1900) with succinic anhydride at 150 °C for 5 h, followed by purification via double precipitation[1].
mPEG-CM (MW 2000) (0.2:1-1:1 feed molar ratio to PCA-g-PCL; 1 h mixed) conjugates with PCA-g-PCL via electrostatic interaction to form spherical polyion complex micelles, where higher mPEG-COOH to PCA-g-PCL molar ratios produce smaller micelles, and mPEG@PCA-g-PCL(1:1) micelles load paclitaxel with 7.8% drug loading and 78.6% encapsulation efficiency[1].
mPEG-CM (MW 2000) (0.2 g starch; 0.38 g mPEG-CM; 0.144 g EDC·HCl; 0.046 g DMAP in 15 mL DMSO; 48 h at 25 °C; 3 days (dialysis)) conjugated to soluble starch via EDC·HCl/DMAP coupling in DMSO at 25 °C for 48 h produces mPEGylated starch (mPEG-St) with a 76.4% yield[2].
