N-Demethyl amiton oxalic
N-Demethyl amiton oxalic is an irreversible cholinesterase (ChE) inhibitor. N-Demethyl amiton oxalic elicits low-level postganglionic neuronal firing in cat vesical parasympathetic ganglia by preventing the breakdown of endogenously released ChE. N-Demethyl amiton oxalic inhibits ChE activity in squid optic ganglia and induces hyperactivity, ink ejection, pigment changes, and death, with a rate of enzymatic detoxification much lower than that of DFP. N-Demethyl amiton oxalic can be used in studies related to the mechanisms of cholinergic transmission in parasympathetic ganglia and neurotoxicology.

Biological Activity
Description
In Vivo
N-Demethyl amiton oxalic (compound 217AO) (100-500 μg/kg; intra-arterial injection; single dose) induces low-level postganglionic neuronal discharge in the cat vesical parasympathetic ganglia, an effect that is blocked by Atropine (HY-B1205)[2].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model:Loligo pealei (either sex, 15-20 cm in length, 50-100 g in weight)[1]
-
Dosage:1.0 mg/kg; 2.0 mg/kg; 3.0 mg/kg; 4.0 mg/kg
-
Administration:i.o.
-
Result:Reduced optic ganglia ChE activity to 30.8% of control levels at 1.0 mg/kg, with 0 of 6 squids dead.
Reduced optic ganglia ChE activity to 12.3% of control levels at 2.0 mg/kg, with 3 of 5 squids dead.
Reduced optic ganglia ChE activity to 8.2% of control levels at 3.0 mg/kg, with 4 of 4 squids dead.
Reduced optic ganglia ChE activity to 4.9% of control levels at 4.0 mg/kg, with 4 of 4 squids dead.
Yielded an approximate LD50 of 1-2 mg/kg.
Caused ≥90% reduction in optic ganglia ChE activity at lethal doses relative to untreated squids.
