NG-nitro-L-arginine

NG-nitro-L-arginine (Nω-nitro-L-arginine; L-NOARG; L-NG-Nitroarginine) is a nitric oxide synthase (NO Synthase) inhibitor, with a Kd of 15 nM for constitutive NOS from bovine brain and a Ki of 4.4 μM for inducible NOS from mouse macrophages. NG-nitro-L-arginine competitively blocks the synthesis of NO from L-arginine; in addition, it inhibits endothelium-dependent vasodilation, and exhibits effects such as blood pressure elevation, heart rate reduction and vasoconstriction. NG-nitro-L-arginine regulates mitochondrial respiratory efficiency, antioxidant enzyme activity, lipid peroxidation level and tissue lead content in rats. NG-nitro-L-arginine can be used in studies related to cardiovascular diseases.

NG-nitro-L-arginine (L-NG-Nitroarginine) acts as a rapidly reversible competitive inhibitor of iNOS in mouse macrophages, with a Ki value of 4.4 μM; it also functions as a slow-onset, high-affinity binding inhibitor of cNOS from bovine brain, with a Kd value of 15 nM, and L-arginine competes with NA for binding to the active site of cNOS[3].
NG-nitro-L-arginine potently inhibits NADPH oxidation catalyzed by bovine brain cNOS, with an association rate constant of 3.2 × 104 M-1s-1[3].
NG-nitro-L-arginine dissociates completely from inhibited bovine brain cNOS upon reactivation with saturating concentrations of L-arginine[3].

NG-nitro-L-arginine (L-NNA) (35 mg/kg, i.p., 30 d pre/post lead) produces divergent hepatic effects in lead-poisoned rats based on hypoxia tolerance: it reduces liver lead only in low-tolerance rats post exposure, increases hepatic nitrate solely in high-tolerance rats, impairs NADH-dependent mitochondrial respiration in low-tolerance rats while restoring succinate-linked respiration in high-tolerance rats, exacerbates protein oxidation in low-tolerance rats, mitigates lipid peroxidation in high-tolerance rats, and elevates SOD/GR/GPx but suppresses CAT in high-tolerance rats[2].
