Phenylalanylalanine (Phe-ala) is an enkephalinase inhibitor and belongs to the class of dipeptides. Phenylalanylalanine inhibits dipeptidyl carboxypeptidase-mediated enkephalin degradation by cleaving the Gly3-Phe4 bond, and when co-administered with Leu-enkephalin, it produces analgesic effects with a synergistic enhancement, which can be reversed by Naloxone (HY-17417A). Phenylalanylalanine can be used in pain-related research.

Phenylalanylalanine (Phe‑Ala) can undergo solid-state intramolecular cyclization to form cyclo (Phe‑Ala); cyclo (Phe‑Ala) can self-assemble into fibrous structures on the HOPG surface, exhibiting supramolecular gelation properties[2].

Result:Yielded an average response latency standard deviation of 3.1 sec across 13 observations after 250 mg/kg i.p. dosing, triggering analgesic responses exceeding the 45 sec threshold at 3 h and 5 h post‑administration.
Obtained an average response latency standard deviation of 4.3 sec across 14 observations after 12.5 mg/kg i.c.v. dosing, inducing analgesic responses exceeding the 45 sec threshold at 1.5 h, 2 h, 2.5 h and 10 h post‑administration.
Achieved an average response latency standard deviation of 4.6 sec across 14 observations after 12.5 mg/kg i.c.v. co‑administration with 20 mcg Leu‑enkephalin, generating prolonged and enhanced analgesic responses exceeding the 45 sec threshold from 1.5 h to 10 h post‑administration compared with separate i.c.v. injection.
Displayed analgesic effects reversible by naloxone, with latency recovering to baseline within 5-10 min.
