Phosphofructokinase,Bacillus stearothermophilus is a glycolytic enzyme and also an allosteric activation target of fructose-2,6-bisphosphate. PFK1 supports cardiac glycolysis to maintain normal cardiac function and mediates adaptive responses to pressure overload. Upregulated expression and activity of PFK1 are associated with the alleviation of pathological changes induced by pressure overload, including diastolic dysfunction, interstitial fibrosis and myocardial hypertrophy. Inhibition of PFK1 redirects glucose carbon flux to the pentose phosphate pathway. PFK1 promotes proliferation, migration and glycolysis of colorectal cancer cells, reduces apoptosis and decreases their radiosensitivity, and shows higher expression levels in radioresistant tumors. PFK1 can be used in research related to diastolic dysfunction heart failure and colorectal cancer.

Phosphofructokinase,Bacillus stearothermophilus (PFK1) is expressed in a variety of human colorectal cancer (CRC) cell lines, with the highest expression level detected in HT29 cells; in addition, its expression level in radioresistant tumors is significantly higher than that in radiosensitive tumors[3].

TUNEL staining for apoptotic DNA fragmentation
TUNEL staining detects DNA strand breaks by using terminal deoxynucleotidyl transferase to add labeled nucleotides to exposed 3′-OH DNA termini, generating either microscopic staining in fixed cells or tissue sections, or fluorescence/cytometric signal in cell suspensions. TUNEL positivity reflects DNA fragmentation but should not be interpreted alone as definitive apoptosis, because TUNEL can also label necrotic, autolytic, mechanically damaged, or DNA-repair-associated DNA breaks.

Cell Viability Determination by MTT Colorimetric Assay
The following protocol uses the MTT colorimetric assay as a classic literature-established method for assessing cell viability/metabolic activity in cultured mammalian cells. MTT[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] is reduced by metabolically active cells to a colored formazan product; the amount of formazan is quantified spectrophotometrically and provides an indirect measure of metabolically active viable cells. Importantly, MTT reduction reflects cellular oxidoreductase/metabolic activity rather than an absolute direct count of living cells, so changes in cellular metabolism can alter the signal independently of cell number.
