Description

Probe-Cys is a water-soluble and selective near-infrared fluorescent probe for Cysteine (Cys) (λex= 680 nm, λem=710 nm) that is not interfered by Hcy, GSH, and HS-. Probe-Cys can react with the stimulant DTT (HY-15917) and the NEM (HY-D0843) in HepG2 cells and zebrafish for the detection of endogenous Cys. Probe-Cys can also be used for imaging Cys in Arabidopsis thaliana. Probe-Cys provides a method for cancer diagnosis and exploration of plant sulfur metabolism[1].

In Vitro

Probe-Cys sensitively detects Cys in cell-free systems with a linear range of 0-100 μM, a detection limit of 0.035 μM, and a strong fluorescence turn-on response at 710 nm[1].
Probe-Cys (10 μM; 1 h) specifically detects endogenous and exogenous Cys in HepG2 cells, with no interference from Hcy or GSH, and responds to modulations in cellular Cys levels via DTT and NEM treatment[1].
Probe-Cys (10 μM; 1 h) penetrates Arabidopsis thaliana root tissue to detect endogenous Cys, with fluorescence responding to DTT-mediated elevation and NEM-mediated inhibition, and reveals higher Cys content in root meristematic zones compared to elongation zones[1].

In Vivo

Probe-Cys (20 μM; 1 h) exhibits good permeability and detects endogenous and exogenous Cys in healthy zebrafish, with DTT (HY-15917) -pretreated zebrafish showing 96.61% of the fluorescence intensity of Cysteine (HY-Y0337)-pretreated zebrafish[1].

Result: Exhibited weak fluorescence with relative pixel intensity ca.
32.62% (defining the Cys-pretreated group intensity as 1.0).
Showed greatly reduced fluorescence with relative pixel intensity ca. 7.54% when zebrafish were preincubated with NEM (HY-D0843).
Showed obvious fluorescence enhancement with relative pixel intensity ca. 96.61% when zebrafish were preincubated with DTT.
Produced stronger fluorescence signals in the yolk sac compared to other areas.
