PROTAC JAK1 degrader 1 (Compound 10c) (0.12-10 μM; 0-24 h) selectively degrades JAK1 in Hel cells in a time- and concentration-dependent, proteasome-dependent manner with a DC50 of 214 nM, while sparing JAK2, JAK3, and TYK2, and suppresses downstream JAK-STAT signaling via JAK1 degradation[1].
PROTAC JAK1 degrader 1 potently inhibits proliferation of Hel, THP-1, and K562 cells with IC50 values of 1.3 nM, 6.1 nM, and 10.5 nM, respectively[1].

Result:Induced 79.5% degradation of JAK1 and only 16.0% degradation of JAK2 at 10 μM for 24 h.
Induced > 70% JAK1 degradation within 8 h at 5 μM, with time-dependent degradation observed through 24 h.
Induced concentration-dependent JAK1 degradation with a DC50 of 214 nM, and failed to induce significant degradation of JAK2, JAK3, or TYK2 at tested concentrations.
Blocked JAK1 degradation was observed when cells were pretreated with MG132 (10 μM), MLN4924 (100 nM), or carfilzomib (100 nM) prior to 5 μM compound treatment for 24 h.
Induced JAK1 degradation and suppressed phosphorylation of STAT1 and STAT3 at 1 μM for 24 h; this suppression was attenuated by co-treatment with MG132 (10 μM).

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (3.16 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
