DYRK2

DYRK2 is a nuclear DYRK-family kinase with an established role in genome-stability signaling. Upon DNA damage it phosphorylates p53 at Ser46, a modification that drives pro-apoptotic gene expression, and it also phosphorylates the substrate-recognition protein SPOP to control turnover of targets such as c-Myc and c-Jun. DYRK2 associates with complexes implicated in meiotic chromosome dynamics and DNA repair, and its own stability is regulated in a damage-dependent manner. Expression is broad but modest compared with DYRK1A, and reduced DYRK2 activity has been linked to chemoresistance. Recombinant DYRK2 enables p53-pathway studies, substrate validation, and screening of DYRK-family inhibitors.
