Description

REF1 peptide is a PORK1 agonist with an EC50 of 0.028 nM in tomato. REF1 peptide binds to the extracellular domain of PORK1, triggers receptor autophosphorylation, and drives downstream MPK3/MPK6 activation, thereby initiating defense and regeneration signaling cascades. REF1 peptide induces callus formation, inhibits shoot regeneration upon continuous exposure, and enhances the regeneration and transformation efficiency of recalcitrant dicotyledonous and monocotyledonous crops[1][2].

In Vitro

REF1 peptide (100 nM; 12 h) potently induces the expression of the PI-II defense gene in wild-type Solanum lycopersicum seedlings[1].
REF1 peptide (1 nM; 7-14 d) restores the callus formation and shoot regeneration defects of hypocotyl explants from Solanum lycopersicum prp-2, prp-3 and spr9 mutants[1].
REF1 peptide (100 nM; 30 min) induces the activation of MPK3/6 in Solanum lycopersicum seedlings in a PORK1 receptor-dependent manner[1].
REF1 peptide (2 μg/mL; 24 h) increases the p53-mediated transcriptional activation levels of p21, Bax and GADD45 promoters by approximately 2-fold in H1299 cells[2].
REF1 peptide (20 μg/10-cm plate; 12-72 h) increases p53-induced endogenous p21 mRNA levels and elevates cyclin G mRNA levels in H1299 cells[2].

Incubation Time: 72 h (transfection); 12 h (tetracycline maintenance/removal)
Result: Further increased p53-induced p21 mRNA by ~40%.
Enhanced p53-induced cyclin G mRNA increase (2-fold without REF1 peptide) by 50-100%.
Had no effect on p21 or cyclin G mRNA in the absence of p53.
