San45 is a nonselective dual amylin and calcitonin receptor agonist (DACRA) with nonselective potency relative to calcitonin receptor (CTR) and amylin 1 receptor (AMY₁R), and acts as a stabilizer of receptor-ligand complex and prolonged activator of cAMP signaling. San45 carries a conjugated lipid modification that can be used for the research of obesity.

Biological Activity
San45 (serial dilutions; 30 min) is a potent, nonselective dual amylin and calcitonin receptor agonist (DACRA) with similar potency at AMY3R (pEC50 = 10.66) and CTR (pEC50 = 10.45) in HEK293-Flp-In cells[1].
San45 (serial dilutions; 30 min) maintains potent, nonselective DACRA activity across CTR, AMY1R, and AMY3R in COS-1 cells, with pEC50 values of 9.19 (CTR), 8.83 (AMY1R), and 9.34 (AMY3R)[1].
San45 (4 μM (AMY3R complex); 2 μM (CTR complex)) binds to AMY3R via a distorted bypass conformation and to CTR via an alternative CT-like conformation, stabilized by its lipid modification, supporting its nonselective DACRA activity[1].
San45 (serial dilutions; measured over cycles) exhibits prolonged cAMP signaling duration in COS-7 cells expressing AMY1R, likely due to its lipid modification[1].
