Biological Activity
CLOCK[1]
CLK8 (10-40 μM; 4-6 d) enhances the amplitude of the Bmal1-dLuc signal in U2OS and NIH 3T3 cells in a dose-dependent manner with no period change[1].
CLK8 (10-40 μM) reduces BMAL1-CLOCK interaction, whereas the interaction between CLOCK-F80A, K220A and BMAL1 is not affected in HEK293T cells[1].
CLK8 (20 μM; 2 d) reduces nuclear localization of CLOCK in U2OS cells[1].

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
