SppIP (Sakacin P inducer peptide) is a peptide consisting of 19 amino acids. SppIP is an essential inducer for sakacin P production. SppIP can induce the expression of PRRSV GP5 and SARS-CoV-2 spike protein in recombinant Lactobacillus plantarum. SppIP can activate the transcription of the sakacin P promoter to drive the expression of downstream heterologous capsid proteins, increasing the proportion of PCV2d capsid proteins displayed on the cell surface of Lactobacillus plantarum. SppIP can be used in studies related to sakacin P induction.

Description

SppIP (Sakacin P inducer peptide) is a peptide consisting of 19 amino acids. SppIP is an essential inducer for sakacin P production. SppIP can induce the expression of PRRSV GP5 and SARS-CoV-2 spike protein in recombinant Lactobacillus plantarum. SppIP can activate the transcription of the sakacin P promoter to drive the expression of downstream heterologous capsid proteins, increasing the proportion of PCV2d capsid proteins displayed on the cell surface of Lactobacillus plantarum. SppIP can be used in studies related to sakacin P induction[1][2][3][4].

In Vitro

SppIP (10-200 ng/mL; 4-8 h) induces expression of PRRSV GP5 in recombinant Lactobacillus plantarum CGMCC 1.557, with maximum production achieved using 200 ng/mL SppIP for 7 h at 33 °C in the pSIP-1320-O5MH strain, and expression remains stable across 5 bacterial passages[2].
SppIP (50 ng/mL; 8 h) induces expression of SARS-CoV-2 spike protein in Lactobacillus plantarum Lp18:S that is stable after 20 min at 50 °C or 30 min at pH = 1.5, and shows increased expression after 2 h exposure to 0.2% bile salt[3].
SppIP (50 ng/mL; 3-12 h) induces time-dependent expression and surface localization of PCV2d His6-tag-capsid protein in recombinant Lactobacillus plantarum pLp3050-His6-tag-capsid, with maximal production (4.8 μg per 1010 CFU) in cell wall fractions after 12 h, while also reducing bacterial growth rate[4].
