Succinylacetone pyrrole
Succinylacetone pyrrole is a potent inhibitor of δ-aminolevulinic acid dehydratase (ALA dehydratase) with a Ki value of 5 μM. Succinylacetone pyrrole blocks the biosynthetic pathway from ALA to porphobilinogen (PBG) and subsequent porphyrins or corrinoids by competitively binding to ALA dehydratase and inhibiting its activity. Succinylacetone pyrrole can be used for studies on vitamin B12 biosynthesis and its regulatory mechanisms.

Succinylacetone pyrrole (18 h) inhibits corrinoid synthesis in Clostridium tetanomorphum cells. Treatment with 1.1 μM Succinylacetone pyrrole reduces corrinoid production by 49%, and a concentration of 8.2 μM reduces it by 83%[1].
Succinylacetone pyrrole (2-10 μM) exerts an inhibitory effect on porphyrin formation in Staphylococcus epidermis, with approximately 50% inhibition observed at 4 μM[1].
Succinylacetone pyrrole potently inhibits purified ALA dehydratase from Clostridium tetanomorphum via a competitive binding mechanism, with a Ki value of 5 μM[1].
