Sulfane sulfur probe 4 (SSP4) is a fluorescent indicator with high selectivity and sensitivity toward sulfane sulfur (including small-molecule sulfane sulfur and protein-bound sulfane sulfur), and its fluorescence is inhibited by Cu2+. Sulfane sulfur probe 4 produces a fluorescence turn-on response upon reaction with sulfane sulfur. Sulfane sulfur probe 4 can detect persulfidated proteins including papain, BSA, GAPDH, and human protein disulfide isomerase. Sulfane sulfur probe 4 enables bioimaging of exogenous and endogenous sulfane sulfur in cells.

SSP4 (5 μM; 20 min) is a highly sensitive and highly selective fluorescent probe targeting small-molecule sulfane sulfur in cell-free systems, with a limit of detection of 28 nM. It exhibits optimal performance under pH 6.5-8 conditions and is hardly interfered with by most biomolecules except for high concentrations of GSH and Cu2+[1].
SSP4 (5-20 μM; 20-30 min) enables sensitive detection of exogenous and endogenous sulfane sulfur in HeLa, COS-7 and HEK293T cells. No cytotoxicity is observed at the relevant concentrations, and it can distinguish differences in intracellular sulfane sulfur levels based on genetic modification[1].
