W-GTF01
W-GTF01 is a MGAT1 inhibitor. W-GTF01 blocks the MGAT1-catalyzed glycosylation of CD73, and inhibits CD73 dimerization, membrane translocation and adenosine production. W-GTF01 restores the function of IFNγ-producing CD8+ T cells, enhances tumor infiltration and activation of CD8+ T cells, and suppresses tumor growth. W-GTF01 can be used in research related to triple-negative breast cancer.

Biological Activity
W-GTF01 restores tumor immune response in immunosuppressive breast cancer cells and reduces CD73 membrane localization in MDA-MB468 and MDA-MB231 triple-negative breast cancer (TNBC) cells by blocking MGAT1-mediated CD73 glycosylation[1].
W-GTF01 potently and dose-dependently inhibits MGAT1 enzymatic activity via specific binding to the MGAT1 active site[2].
W-GTF01 revives CD8+ effector T cell function (including IFNγ production) and reduces MDA-MB468 TNBC cell survival in a CD73-dependent manner in 2D coculture with PBMCs[2].
