XL-3156 is a cGAS inhibitor. XL-3156 occupies both allosteric and orthosteric sites simultaneously, and inhibits the interaction and phase separation between cGAS and DNA by stabilizing the closed conformation of the activation loop. XL-3156 can be used in the research of diseases such as autoimmune diseases and inflammation.

Biological Activity
XL-3156 (20 μM; 20 min) inhibits the interaction between purified h-cGASCD and Cy5-labeled ISD45 DNA in EMSA assays[1].
XL-3156 (4-100 μM) dose-dependently inhibits phase separation between purified h-cGASCD and FAM-labeled ISD100 DNA in vitro[1].
XL-3156 (50 μM; 20 min) disrupts condensate formation between purified h-cGASCD and Cy3-labeled ISD100 DNA, with an EC50 of 783 nM[1].
XL-3156 (100 μM; 30 min) significantly inhibits phase separation between purified full-length h-cGASFL and Cy3-labeled ISD100 DNA in vitro[1].
