Guidelines (The following is our recommended protocol, which serves only as a guideline and should be modified according to your specific needs)[1].
1. Preparation of Working Solutions:
Prepare a 60 μM Z-Arg-Arg-AMC hydrochloride solution.
2. Specific Procedures:
1) Prepare a 1:3000 diluted working solution of human cathepsin B in periplasmic analysis buffer (PAB; sodium bicarbonate-free, containing 0.6 mM CaCl2, 0.6 mM MgCl2, 2 mM L-cysteine, 25 mM Pipes, adjusted to pH 7.0), corresponding to an enzyme concentration of 225 ng/mL with a specific activity of 588 U/mg.
2) The enzymatic reaction system has a volume of 0.2 mL and is performed in a 96-well plate. The final enzyme dilution ratio is 1:30000 (22.5 ng/mL; 0.9 mM), with 60 μM Z-Arg-Arg-AMC as the substrate.
3) Inhibitors and their dilutions are prepared with dimethyl sulfoxide. The reaction system of the untreated group receives an equal volume of dimethyl sulfoxide containing excipients (final concentration 0.1% v/v). A Fmax fluorescence microplate reader set to kinetic mode with the accompanying SoftPro software (Molecular Devices, Sunnyvale, CA, USA) measures the reaction rate (relative fluorescence units per minute), with an excitation wavelength of 355 nm and an emission wavelength of 460 nm. The reaction proceeds at 37 ℃ for 10 min, with data read every 30 s.

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (3.80 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (3.80 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
