Zinpyr-1 is a zinc-responsive fluorescent indicator and a membrane-permeable metal-binding probe. Zinpyr-1 forms a complex with Mn2+ ions and generates a fluorescence turn-on signal. Zinpyr-1 binds to free zinc ions in serum, enabling quantitative detection of free zinc concentration. Zinpyr-1 produces fluorescence signals reflecting the relative zinc concentration in plant root cells, localizes to specific layers of plant root cells, and can be used to support analyses related to plant zinc transporter mutations and homeostasis.

    Morselized human allograft was treated with 0.5% (w/v) ZnCl2 dissolved in TBS buffered to pH 7.4 for 60 min and then washed extensively with TBS before detecting bound Zn with the fluorescent stain, Zinpyr-1. The stained allograft morsels were polished to reveal the interior of the bone morsels before epi-fluorescent detection of the Zinpyr-1.     Zinpyr-1 is a zinc-responsive fluorescent indicator and a membrane-permeable metal-binding probe. Zinpyr-1 forms a complex with Mn2+ ions and generates a fluorescence turn-on signal. Zinpyr-1 binds to free zinc ions in serum, enabling quantitative detection of free zinc concentration. Zinpyr-1 produces fluorescence signals reflecting the relative zinc concentration in plant root cells, localizes to specific layers of plant root cells, and can be used to support analyses related to plant zinc transporter mutations and homeostasis[1][2][3].

    In Vitro

    Zinpyr-1 (10 μM; pH 6-8) forms a 1:2 complex with Mn2+. Compared to metal-free Zinpyr-1, this complex exhibits a 25-fold enhanced fluorescence dynamic range for Zn2+ detection (106 at pH 7.0). It also maintains a high dynamic range at pH 6.0 and 8.0, and shows better selectivity for Zn2+ over the abundant monovalent and divalent cations in organisms[1].
    Zinpyr-1 (10 μM) forms a Mn2+ complex that can penetrate cell membranes, exhibits no toxicity to HeLa cells, has extremely low background fluorescence, and shows strong activation-type fluorescence responsive to zinc ions, making it suitable for live-cell imaging[1].
    Zinpyr-1 (0.1 mM) forms a Mn2+ complex that functions as a zinc-responsive MRI contrast agent[1].
    Zinpyr-1 stably induces the minimum fluorescence (Fmin) and maximum fluorescence (Fmax) required for free zinc quantification in serum, with the maximum excitation and emission wavelengths being 507 nm and 526 nm, respectively, enabling reliable quantification of free zinc in serum samples[2].
    Zinpyr-1 (5-20 μM; 15 min-3 h) serves as an effective sensor for relative zinc levels in root cells of Arabidopsis thaliana seedlings, and its fluorescence intensity reflects intracellular zinc concentrations[3].
