4,4-Dimethyl-2-cyclopenten-1-one (4,4-Dimethylcyclopent-2-enone) is a β-unsubstituted cyclic α,β-unsaturated ketone and anticancer agent. 4,4-Dimethyl-2-cyclopenten-1-one does not activate Caspase 3, 8 or 9, nor does it induce internucleosomal DNA fragmentation. 4,4-Dimethyl-2-cyclopenten-1-one exhibits anticancer activity against oral squamous cell carcinoma and promyelocytic leukemia.

Biological Activity
4,4-Dimethyl-2-cyclopenten-1-one (24 h) exhibits selective cytotoxicity against oral squamous cell carcinoma cells HSC-2, HSC-3, HSG and promyelocytic leukemia cells HL-60, but shows no obvious selectivity toward normal gingival fibroblasts HGF, dental pulp cells HPC and periodontal ligament fibroblasts HPLF. Its SI value is 4.0, and the CC50 values of tumor cell lines range from 14.0 to 152 μg/mL[2].
4,4-Dimethyl-2-cyclopenten-1-one (50 μg/mL; 24 h) exhibits cytotoxic activity against promyelocytic leukemia HL-60 cells, while co-treatment with 5 mM N-acetylcysteine (HY-B0215) significantly reduces this activity[2].
4,4-Dimethyl-2-cyclopenten-1-one (5-80 μg/mL; 6 h) does not induce internucleosomal DNA fragmentation in promyelocytic leukemia HL-60 cells even at concentrations as high as 80 μg/mL after 6 h of incubation[2].
4,4-Dimethyl-2-cyclopenten-1-one (5-80 μg/mL; 4 h) does not activate caspase-8 or caspase-9, and only exerts extremely weak or no activation on caspase-3 in promyelocytic leukemia HL-60 cells even when incubated for 4 h at concentrations up to 80 μg/mL[2].

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (22.70 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
