Description

4-Chlorocinnamaldehyde (trans-p-Chlorocinnamaldehyde) is an antiviral agent. 4-Chlorocinnamaldehyde inhibits the replication of Coxsackievirus B3 in cardiomyocytes, but shows poor efficacy in animal experiments. 4-Chlorocinnamaldehyde can be used in studies related to viral myocarditis[1].

In Vitro

4-Chlorocinnamaldehyde (Compound 2) (0.01-1000 μM; 72 h) shows no obvious cytotoxicity in neonatal rat cardiomyocytes, with a CC50 of 6972.47 μM after 72 h of incubation[1].
4-Chlorocinnamaldehyde (0.01-1000 μM; 72 h) weakly inhibits CVB3 replication in neonatal rat cardiomyocytes, with an IC50 of 2109.08 μM and a therapeutic index of 3.31 after 72 h of incubation[1].
4-Chlorocinnamaldehyde (10 μM; 72 h) inhibits the secretion of proinflammatory cytokines TNF-α, IL-1β and IL-6 induced by CVB3 in neonatal rat cardiomyocytes[1].
4-Chlorocinnamaldehyde (10 μM; 12-24 h) inhibits the mRNA and protein expression of proinflammatory cytokines TNF-α, IL-1β and IL-6 in neonatal rat cardiomyocytes induced by CVB3[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

ELISA Assay[1]

Cell Line: CVB3-infected neonatal rat cardiomyocytes
Concentration: 10 μM
Incubation Time: 72 h
Result: Significantly decreased TNF-α, IL-1β, and IL-6 secretion relative to CVB3-infected control cells.

Real Time qPCR[1]

Cell Line: CVB3-infected neonatal rat cardiomyocytes
Concentration: 10 μM
Incubation Time: 12 h

Western Blot Analysis[1]

Cell Line: CVB3-infected neonatal rat cardiomyocytes
Concentration: 10 μM
Incubation Time: 24 h
Result: Significantly reduced TNF-α, IL-1β, and IL-6 protein expression relative to CVB3-infected control cells.

In Vivo

4-Chlorocinnamaldehyde (Compound 2) (20-60 mg/kg; p.o.; daily; 6 days) does not significantly reduce myocardial pathological scores or viral titers in Coxsackievirus B3-induced viral myocarditis in male BALB/c mice[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: BALB/c (four-week-old, male, Coxsackievirus B3-induced viral myocarditis model)[1]
Dosage: 20 mg/kg; 40 mg/kg; 60 mg/kg
Administration: p.o.; daily; 6 days
Result: Showed no statistically significant differences in pathological scores or viral titers between any dose group and the model control group.

In Vivo:

Select the appropriate dissolution method based on your experimental animal and administration route.

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

  • Protocol 1

Add each solvent one by one:  10% DMSO    90% Corn Oil

    Solubility: ≥ 3.33 mg/mL (19.99 mM); Clear solution

This protocol yields a clear solution of ≥ 3.33 mg/mL (saturation unknown). If the continuous dosing period exceeds half a month, please choose this protocol carefully.

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (33.3 mg/mL) to 900 μL Corn oil, and mix evenly.

If the continuous dosing period exceeds half a month, please choose this protocol carefully.
Please ensure that the stock solution in the first step is dissolved to a clear state, and add co-solvents in sequence. You can use ultrasonic heating (ultrasonic cleaner, recommended frequency 20-40 kHz), vortexing, etc. to assist dissolution.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
