Ac-VEID-AMC is a fluorescent substrate for the determination of caspase-6 and related cysteine protease activities (Ex=340-360 nm, Em=440-460 nm)[1].

In Vitro

Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
Enzyme activity (rabbit osteoclasts for example)[1]:
1. Treat purified, differentiated cells by medium for 48 h (time depends on the cell).
2. Put treated cells into 4.5 mL lysis buffer.
3. Each lysate is divided into thirds, then incubate with 40 μM Ac-VEID-AMC at 37°C for 1 h.
4. Detect cleavage of fluorescent substrates (Ex=340-360 nm, Em=440-460 nm)
