Description

AgrC-IN-1 is an AgrC inhibitor with an IC50 of 3.5 μM against Staphylococcus aureus AgrC. AgrC-IN-1 competitively binds to AgrC, inhibiting its autophosphorylation activity in Staphylococcus aureus. AgrC-IN-1 inhibits quorum sensing in Staphylococcus aureus, blocking virulence factor production. AgrC-IN-1 can be used for the research of Staphylococcus aureus infections[1].

In Vitro

AgrC-IN-1 (CP-20) (16 μM) completely inhibits P3 transcription in the agr-null background Staphylococcus aureus group-I strain with isolated AgrC/A signal transduction system, strongly suggesting it targets AgrC or AgrA[1].
AgrC-IN-1 has no significant effect on the luminescence activity mediated by AgrA:DNA binding in the Staphylococcus aureus strain with agr operon replaced by luciferase reporter[1].
AgrC-IN-1 causes a dose-dependent decrease in AgrC autophosphorylation level when co-incubated with AIP-I in the Staphylococcus aureus AgrC nanodisc system, directly inhibiting AgrC autophosphorylation[1].
AgrC-IN-1 (10 μM) displaces 375 nM TAMRA-AIP-III D4A from Staphylococcus aureus AgrC nanodiscs and reduces the fluorescence polarization signal by 52%, indicating its binding to AgrC and competitive occupation of the binding site[1].
